Melinda Gordon is a young woman from the fictional town of Grandview, New York who has the ability to see and communicate with the dead. Melinda lives with her husband Jim Clancy. She is the owner of an antique shop called “Same as it Never Was.” Ghosts seek Melinda’s help in relaying a message or completing a task that will put the spirit to rest, and allow them to cross over into The Light (which is possibly Heaven). Those who died with unfinished business become earth-bound and can not cross over, and Melinda, as their earthly representative, helps them to find peace. The show does not present the ghosts as having sinned; rather it is the spirits’ own guilt that condemns them, and their own fear of judgement that keeps them from “crossing over” into an afterlife.The first season introduces Romano, a former cult leader from Europe who influenced his followers to commit a mass suicide in 1939. His own suicide transformed him into an earth-bound negative entity. Romano attempts to do the exact opposite of Melinda and gather earth-bound souls and prevent them from crossing over into The Light. Melinda and Romano struggle over the 300 souls of people who die in a airplane crash in Grandview. Melinda also suffers a huge personal loss at the end of the season as her best friend, Andrea (Aisha Tyler), dies when her car is caught in the path of the crashing plane.In the second season, Melinda meets Delia Banks, her son Ned Banks, and Professor Rick Payne, all of whom, by season’s end, come to learn of Melinda’s ability, with varying reactions. Delia remains slightly skeptical.

The second season revolves around the thinning veil between the living and the dead. Melinda meets Gabriel, who has similar abilities to hers. But Gabriel is working for the Shadows (as yet unnamed) and working to make sure that ghosts do not cross over into The Light, and in fact is working to block The Light from all ghosts.

In the third season, Melinda searches in her family history for answers as she gets closer and closer to learning the secret of her gift, her childhood, and her father. Melinda also finds out Gabriel is her half-brother. He is the son of Tom Gordon (Melinda’s father).

In the fourth season, Melinda meets Eli James after a fire at Rockland University and who, after his own near-death experience, develops the ability to listen to ghosts. Melinda says goodbye to her close friend Rick Payne, who leaves Grandview on a research trip for the university. In this season, Jim is shot and killed. He does not “cross over” because he does not want to leave Melinda, and his spirit later enters the body of a man who died in an accident. When the man regains consciousness, he has no memory of being Jim. Melinda works to get him to remember, and succeeds after much difficulty and skepticism on the part of her friends.In the fifth season, we are introduced to Jim and Melinda’s son, Aiden Lucas. Melinda sometimes worries about her son, because he is destined to be more powerful than she is. He can see the “Shinies and the Shadows.” Melinda, Ned, and Eli, with the frequent help of Delia, work to keep the Book of Changes, a mystical object, safe from the Shadows.

A spotlight on the StAR protein

Commonly shortened to StAR, the steroidogenic acute regulatory protein plays a key role in mitochondrial transport within steroid-producing cells, regulating hormone production by controlling cholesterol transfer within the mitochondria. StAR is a member of the START domain protein family, and is also known as START domain-containing protein 1, or STARD1. Antibody suppliers may list it under either or both names. Star antibodies have proven important in several areas of research.

StAR is a protein of 285 amino acids, located on chromosome 8p. It is found in a number of steroid-producing areas including the ovary, testes and adrenal cortex. Its primary purpose is to mediate transfer of cholesterol from the outer mitochondrial membrane to the inner surface, so that the enzyme cytochrome P450scc can cleave the side chain, performing the first stage of steroid synthesis.

Cholesterol is a lipophilic protein and is therefore unable to cross the aqueous membrane layer unaided. Antibody studies have shown StAR to be the most important mediator of this process, assisted by a number of other proteins. These include TSPO, SCP2 and the Steroidogenic Activator Polypeptide SAP.

It is known that the signalling sequence by which StAR is targeted to the mitochondria is a two-stage process, with activity increased by phosphorylation at position 195. Cholesterol transfer is achieved via the START (StAR-related transfer) domain. However, the exact mechanism of action by which StAR achieves cholesterol transport remains unclear. A number of mechanisms have been suggested, including binding to cholesterol in a shuttle-type process; however, the evidence points to it acting more like a channel. Other hypotheses include desorption of the cholesterol through the membrane; the creation of contact sites, or interaction with PBR.

StAR expression may not be as restricted as we think. Recently, J.M Taylor and others published a study showing StAR expression in macrophages, where it stimulated production of 27-hydroxycholesterol, suggesting a possible anti-inflammatory role in cardiovascular disease. An earlier study suggested a role in bile production. Although neither of these studies has proven conclusive, it seems the StAR antibodies we at Novus Biologicals have on our antibody database have more secrets yet to unveil.

From War Zone to Franchise Zone

Ross Paterson has found the perfect spot for would-be entrepreneurs: The place has minimal market penetration, abounds in hardworking, business-savvy people and has minimal government red tape. Last spring, the former Army officer took his Fort Worth, Texas-based Growth Coach business-consulting franchise to Afghanistan for a week of seminars to teach 30 nascent entrepreneurs at Kabul’s Leadership Development Institute about good ol’ American-style bootstrapping.

How does a Texas business coach set up shop in Afghanistan?

I went over there for the first time in 2002 as part of a nonprofit I run called Global Fusion, and now I go back a few times a year. The idea is to create a vision and then get the resources to bring about a new Afghanistan–like working with universities to create an agricultural model that moves away from opium and educating women on how to keep their families healthy. The more we got involved, the more we realized we needed to make everything into a business model so it could be sustainable there. I realized the material I used in the Growth Coach franchise I started in 2006 could give them the systematic model they need.

Isn’t business in Afghanistan a little more intense than here?

They are very business-oriented people, they just work in a different system than us; it’s very tribal, and very honor- and shame-oriented. Big debacles only happen if you don’t learn what’s culturally relevant. But Afghan entrepreneurs need the exact same things small-business owners in America need: focus, alignment, leadership, and good systems and processes. I had cell phone entrepreneurs, doctors, newspaper owners and eight women in my class. Out in the rural edges, it gets more extreme, but the people at my seminar have the same hopes and dreams you have for peace and prosperity.

Was your franchisor on board with your trip?

Oh yeah. The whole Growth Coach company has a passion to make a difference. When I told them what I was doing, they made financial commitments and provided training materials.

Will you keep coaching in Kabul?

We were really only able to scratch the surface; they told me they want help with project management, dealing with conflict resolution, leadership in crisis situations and other topics. They’ve invited me back in the fall, and now I’m looking for entrepreneurial experts adventurous enough to go with me.

War’s Review : Microsoft, Facebook could use Skype against Google

Facebook may not have bought Skype, but the social networking company may still reap the benefits.

Earlier this week, Microsoft announced that it had reached a deal to buy Skype to buy the voice and video communications company for US$ 8.5 billion in cash.

In a press conference Tuesday, Microsoft chief executive Steve Ballmer said the software maker will connect Skype to major Microsoft services, like the Outlook email client and its Xbox and Kinect gaming systems.

Industry analysts, however, surmise that Microsoft Dell inspiron 1520 battery may be willing to go outside its corporate walls and connect Skype with online partner Facebook.

“Facebook could get a little goodness out of the Skype deal, given its close relationship with Microsoft,” said Dan Olds, an analyst with The Gabriel Consulting Group. “I can see where Skype could play a role between them and provide benefits to both sides. Adding Skype-like capabilities to Facebook would give Microsoft a ready-made marketing platform for it and give Facebook a new tool to offer its users.”

Microsoft and Facebook announced last October that they were working together to make search more social. Both companies made it clear at the time that the move was an extension of their ongoing relationship. Qi Lu, president of Microsoft’s Online Services division, said in October that with the pairing, users should expect more tools and technologies from the companies in the future.

Before Microsoft announced that it was buying Skype, most online speculation had Google and Facebook Hp elitebook 8530w battery , which have increasingly competitors, both vying to scoop up Skype. During Tuesday’s press conference, Skype CEO Tony Bates did not answer when a reporter asked him what other companies had shown interest in buying the video communications company.

Rob Enderle, an analyst with the Enderle Group, said Facebook may have lucked out with the Microsoft-Skype deal.

Instead of having to pay a hefty price to get Skype on its own, Facebook may be able to work out a much less expensive deal with Microsoft to use Skype communication tools on its site.

And considering the fact that both Microsoft and Facebook are engaged in a heated competition with Google, it would behoove Microsoft to help its partner take on its biggest foe.

 “If anybody, Microsoft would rather Facebook Hp dv6000 battery have Skype capabilities,” Enderle said. “It wouldn’t surprise me if Microsoft offered this up, treating Facebook more like a partner… Better to take on Google together.”

While Microsoft and Facebook would like to make it tougher on Google, Facebook could also benefit from gaining some Skype services for its users.

 “If Facebook can strike this kind of deal with Microsoft, it could give them the best of both worlds. They’d have the ability to integrate Skype into Facebook without having to buy or manage it,” Olds said. “This would make Facebook stickier by keeping users on the site for a longer period of time, which adds up to more advertising impressions, which is the name of the game.”
